Menu Fully-Spectral Electron Diffraction Analysis of Honey. In the last post, we looked at how to measure the thickness of a honey sample (from a honeycomb cell) using a focused electron beam in a scanning electron microscope. The other important characteristics of honey samples, though, are their grain structure (dendrites, truffles, etc.) and their color. In this post, I’ll show how to determine the element composition of the honey by looking at the background in scanned diffraction images. Figure 2. Measuring the thickness of honey using diffraction. In this example, the sample was a very light one (a honey mixture of honey and grapes), and the diffraction was in the very weak diffraction regime (lower resolution on the detector than the electron beam diameter). So, the background color isn’t affected. In order to improve the statistics, we can slice the image with a single pixel width, and integrate the intensities of the pixels in the slice (Figure 2b). The log-normal integral background of the slice is then subtracted from the original image (Figure 2c). This yields the normalized integrated slice, which can then be compared to the known spectrum of the sample to determine the ratio of the elements. The sum of all the elements in a sample is 100%, and a sample containing say, 10% Co and 90% Cu would be shown as 10% Co and 90% Cu. Figure 3. However, the other elements also appear in the background spectrum, and, by determining the position of the peaks in the slice, we can determine the ratio of these elements
